As part of the ‘Stamets Stack’, niacin is typically introduced after the first 4-week cycle of microdosing.
Niacin is a vitamin that is crucial in various bodily functions is a part of energy and protein metabolism and is known for its antioxidant properties and vasodilation effects (widening of the arteries).
The Stamets Stack protocol recommends higher doses of niacin (50-200mg) than a typical daily supplement (~15mg). When taking higher doses of niacin, the vasodilation effects become more noticeable, leading to what’s known as “flushing”. Flushing refers to the skin developing a temporary rash, which can feel similar to a sunburn—itchy, tingly, red, slightly clammy, and uncomfortable. This typically only lasts for 10-30 minutes and is believed to help in the distribution of the mushroom compounds.

This is a very basic chart that demonstrates a marked increase in the binding affinity of TrkC receptors (which is directly linked to the potential for neuronal stimulation)
There are non-flushing forms of niacin such as niacinamide, which make the ingestion of B3 much more comfortable. However, in the case of the Stamet’s Stack, it’s the flush we are after. The discomfort is part of the practice and (believe it or not) can actually become a pleasant sensation if you manage to focus on the benefits.
Prior to adding Niacin to your microdosing practice, it’s important to rule out any contraindications such as liver disease, kidney disease or stomach ulcers. If you’re not sure, please consult your family doctor prior to beginning the protocol.

Here are some guidelines to help your introduction to Niacin go smoothly:
- Niacin can be purchased at a health food store, or online. Here is a link to the niacin I use.
- Begin with a low dose of niacin (around 50 mg) and gradually increase it daily to induce flushing.
- Your body quickly builds a tolerance to niacin, so you may need to raise the dosage each time you follow the protocol to achieve the flushing effect.
- Take your ‘goldilocks dose‘ + lion’s mane on an empty stomach. Wait 20 minutes, before taking niacin.
- About 5-30 minutes after taking niacin, you’ll start to feel the flush. Don’t worry—this is a normal part of the process. Try to relax and embrace the experience as much as possible.
- Niacin B3 at these dosages is NOT recommended for more than 4 weeks at one time.
- This protocol is only to be undertaken for a maximum of 4 weeks in total. Then, take a break from Niacin for at least 6-8 weeks
